Glucosamine Activates T Lymphocytes in Healthy Individuals and may Induce GVHD/GVL in Stem Cell Transplanted Recipients

چکیده
Graft versus host disease (GVHD) is the major limiting factor after Hematopoietic stem cell transplantation (HSCT). In this study, we report a HSCT-patient who developed arthritis 10-month after allogeneic sibling transplantation, treated with glucosamine-hydrochloride and developed severe acute GVHD within three weeks after the administration of glucosamine. Another HSCT-recipient with an increase in CD33+cells received one dose of donor lymphocyte infusion (DLI). Due to the lack of DLI and based on our experience from the first case, the patient was treated with glucosamine. No sign of relapse was observed in the second patient despite increased number of CD33+ for more than three years. The glucosamine effect was evaluated in seven individuals given the drug for four weeks. The effect of glucosamine treatment was examined by mixed-lymphocyte-reaction (MLR) and the levels of soluble IL-2 receptor (sIL2R), TNF-α and IFN-γ were determined. Glucosamine administration exhibited an enhancement in the allogeneic MLR and an increase in the serum levels of sIL-2R, but a decrease in the serum levels of inflammatory cytokines TNF-α and

Allogeneic disparities in immunoglobuline-like transcript 5 (ILT5) induce potent antibody responses in hematopoietic stem cell transplant recipients Short title: ILT5 specific antibodies in HSCT recipients
In hematopoietic stem cell transplant (HSCT) recipients the recognition of polymorphic antigens by the donor-derived immune system is an important mechanism underlying both graft-versus-host disease (GvHD) and graft-versus leukaemia (GvL) effect. Here we show that a subset of hematopoietic stem cell transplant recipients (13.9 %, n=108) have antibodies directed to surface molecules of dendritic...

Graft-versus-host disease (GVHD) remains a major cause of morbidity and mortality in allogeneic hematopoietic stem cell transplantation. Migration of donor-derived T cells into GVHD target organs plays an essential role in the development of GVHD. beta2 integrins are critically important for leukocyte extravasation through vascular endothelia and for T-cell activation. We asked whether CD18-def...

Despite the presence of toll like receptor (TLR) expression in conventional TCRαβ T cells, the direct role of TLR signaling via myeloid differentiation factor 88 (MyD88) within T lymphocytes on graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) effect after allogeneic stem cell transplantation (allo-SCT) remains unknown. In the allo-SCT model of C57BL/6 (H-2(b)) → B6D2F1 (H-2(b/d)...

Allogeneic hematopietic stem cell transplantation (aHSCT) is widely used for the treatment of hematologic malignancies. Although aHSCT provides a good response against the malignant cells (graft-versus-leukemia [GVL]), it also leads to the development of graft-versus-host disease (GVHD), a severe disease with high mortality and morbidity rates. Therapy for GVHD is commonly based on nonspecific ...
متن کاملSuicide Gene Therapy to Increase the Safety of Chimeric Antigen Receptor-Redirected T Lymphocytes
Chimeric antigen receptors (CARs) are generated by fusing the antigen-binding motif of a monoclonal antibody (mAb) with the signal transduction machinery of the T-cell receptor (TCR). The genetic modification of T lymphocytes with chimeric receptors specific for tumor-associated antigens (TAAs) allows for the redirection towards tumor cells. Clinical experience with CAR-redirected T cells sugge...
